产品介绍:Anti-Human PD-1 Reference Antibody (Daxokafusp Alfa, RUO)|HS870606 是艾必赛生物(abinScience)推出的人源化单克隆参照抗体(人 IgG4-kappa),靶向人程序性死亡蛋白 1(PD-1/CD279),可作为 PD-1/PD-L1 免疫检查点研究的阳性/药物参照质控工具。该抗体在哺乳动物细胞中表达,糖基化更接近天然构象,经 Protein A/G 纯化,纯度 >95%(SDS-PAGE),以液体形式保存在 0.01M PBS, pH 7.4 中;频繁使用 2–8°C,长期建议 -20°C 至 -80°C(自收货日起 ≤12 个月)避反复冻融。(RUO,仅限科研/非临床用途)
PD-1(CD279) 是 B7/CD28 家族的Ⅰ型跨膜抑制性受体,主要表达于活化的 T 细胞、B 细胞、NK 细胞及树突状细胞等,通过与配体 PD-L1(CD274)或 PD-L2(CD273) 结合传递负向共刺激信号,抑制 T 细胞活化与细胞因子分泌,是维持外周耐受、防止自身免疫的关键“刹车”。肿瘤微环境广泛利用 PD-1/PD-L1 轴实现免疫逃逸——肿瘤细胞高表达 PD-L1 与 T 细胞表面的 PD-1 结合,使肿瘤浸润 T 细胞功能耗竭;因此,阻断 PD-1 通路的单抗(如 Pembrolizumab、Nivolumab 等)已成为肿瘤免疫治疗最成功的药物类别之一,在黑色素瘤、非小细胞肺癌、霍奇金淋巴瘤等多种恶性肿瘤中实现持久应答。Daxokafusp Alfa 作为 PD-1 靶点的临床阶段参考分子,其配套参照抗体 HS870606 适用于 ELISA、FACS、体外功能 assay 及体内研究,可用于 PK/PD 研究、方法开发及抗药抗体(ADA)检测等科研场景。
